If that doesn't work, get a support ticket started with Asus right away. With removing the cable to the PCI-E power supply a few times I'm pretty sure you'd have made a good connection. Are you confident your PSU is supplying power to the card?
Also double check the PCI-E Lane switch is on. Kinda weird that it would just stop working suddenly though. Maybe somebody else has ideas.

When you tried to boot what code is the Hex display showing?? This will help us ascertain the nature of the problem. Have you tried resetting bios and booting with only one stick of RAM, furthest socket from CPU with only graphics, keyboard and mouse attached to the system.
You could also try switching to the secondary bios and having a go at booting using the same procedure.
With one card you should use either slot 1 or the third red from cpu down the board.
