I bought a Vantec NexStar JX HDD Enclosure with eSATA (NST-358SU3-BK) to use for a backup drive. I connected it with a back-panel feed-through to the 4th SATA connector on the motherboard. Is it safe to turn this on and off while the computer is on?

es, the ASUS ROG Strix B365-F motherboard supports hot swapping for SATA drives, but you’ll need to enable the option in the BIOS.
To do this:
Enter BIOS: Restart your computer and press the Delete key during boot.
Enable Hot Swap: Navigate to Advanced settings spike, find SATA Configuration, and enable hot swapping for the port your drive is connected to (the 4th connector in your case).
Once this is set up, it should be safe to turn your eSATA drive on and off while the computer is running. Just remember to safely eject the drive in your OS before turning it off to prevent data loss.
