Hi,
For those who have this problem and play RIOT Games, their anti-cheat software “Vanguard” neutralizes something in /System/Device Manager.
Solution :
Disable Vanguard at startup and reinstall realtek drivers.
Since you can't play riot games without vanguard, the problem will return.

What is important is not to update the drivers (because in this case SSVM device/driver remains in place and so it will still disturbing/triggering Vanguard anti-cheat from Riot), but to follow your CLEANUP process as previously indicated in my previous post in order to uninstall SSVM device and delete its driver which will no longer be used. In addition, check that SSVM device is actually uninstalled (even in your hidden devices) : Press Windows key + X > "Device Manager" > "View" > Check "Show hidden devices" > "Sound, video and game controllers" category > Uninstall the "Sonic Studio Virtual Mixer" if you find it in your hidden devices by right-click "Uninstall device" then restart your PC.
3) Follow scrupulously your INSTALL process (from your ASUS ROG SS3 & SS3|DTS Sound Unbound motherboards category) : [DRIVERS] Realtek Audio (AMD 3xx/4xx/5xx/6xx/8xx & Intel 2xx/3xx/4xx/5xx/6xx/7xx/8xx).
For the next drivers packages that I would release, you will can directly follow your UPDATE process (from your ASUS ROG SS3 & SS3|DTS Sound Unbound motherboards category) : [DRIVERS] Realtek Audio (AMD 3xx/4xx/5xx/6xx/8xx & Intel 2xx/3xx/4xx/5xx/6xx/7xx/8xx).

If an ASUS tool (GPUTweak in your case) is disturbing/triggering Vanguard anti-cheat from Riot, only ASUS staff can answer about this.

It's is only the SSVM (Sonic Studio Virtual Mixer) device that is disturbing/triggering Vanguard anti-cheat from Riot, and therefore it is no longer installed since the latest update of the SS3 Extension Driver (for information : https://rog-forum.asus.com/t5/downloads-for-motherboards/drivers-realtek-audio-amd-3xx-4xx-5xx-6xx-8...) :

Finaly not work, vanguard neutralized again.
