I have an ASUS ROG STRIX B650E-E with the latest Realtek Audio Driver and Realtek Audio Console installed (through Armoury crate).
I want to retask the rear line-out connector to headphones but the Realtek Audio Console shows no options to do this even though online help articles say it's possible to.

According to your board's technical specification page at https://rog.asus.com/motherboards/rog-strix/rog-strix-b650e-e-gaming-wifi-model/spec/
My board (ROG MAXIMUS Z790 DARK HERO) has basically similar specification and here's my experience.
Supports: Jack-detection, Multi-streaming, Front Panel Jack-retasking
MULT-STREAMING basically means that you can technically play one sound stream from the front panel connection and a different sound stream from the rear connectors.
FRONT PANEL JACK-RETASKING basically means you can select if the input audio should be treated as MIC level or LINE-IN level. When you connect a microphone the driver should be asking you to specific what have you plugged into the PC. If this prompt is not coming this means in the Realtek Audio Control you have disabled this popup. This is the ONLY re-tasking that is supported.
You can also read my very detailed post https://rog-forum.asus.com/t5/gaming-motherboards/can-asus-support-fix-realtek-audio-drivers-with-bo... which explains that ONLY your front panel connector comes with the headphone amplifier (in your case Savitech SV3H712 AMP).
The audio driver for your board will ONLY allow you to use AMPLIFY to increase the sound volume of the REAR connected device. The Front Panel connected device volume is NOT affected with AMPLIFY which is all mentioned in my post.
I am really not sure what you are trying to do with both Front and Rear devices connected at the same time. I do not think you trying to connect and setup 7.1 speaker setup because I do not see all the connectors in use in your screenshot above.

Unfortunately I can't find the link to it anymore. Are you aware if this is possible to do and if yes how should I configure the connectors?
Even though I couldn't find anyway of retasking the connectors I was able to make the audio louder by changing Realtek Audio Console > Speakers > Amplify level.
