I have a gaming PC connected to the Monitor through DP and Keyboard and mouse are connected to the monitor and all works great.
I have a work laptop that is connected through Lightning Port to USBC on monitor. I have no display output but the Keyboard and mouse will work through the built in hub and it powers my device all day.
I have tried:
This leaves me to think its an issue with the port itself being fault. Can someone advise if I have this right and should raise an RMA?

It was completely my error. I didn't realise that not all USB C cables are created equal and although the one I was using was of high quality, it didn't have capability for 4k60hz. I bought one from UGreen for about £16 that did the job!

Question, does the display for the laptop work at all?
There is a very common issue where devices lose display output at random https://rog-forum.asus.com/t5/gaming-monitors/pg32ucdm-black-screen-during-game-sessions/m-p/1077623...
Asus still has not addressed that issue after almost a year now.
If the laptop just does not have any display output at all, you may have to go into the monitor settings and manually set the display input to usb-c
