Does the m.2_1 slot share lanes with pcie_1 slot?

Open GPUZ 'Bus Interface' click the '?' a window will open click 'start render test'...

1.it should show PCIe X16 4.0 @X16 (1.1) once you start render test it should goto (4.0).

2.If not then if you have anything in your other PCIe slots that might reduce it down to 8x ..

3.Check BIOS that it has not been set to 8x OR 4x... (change it to 'AUTO').

It does, yes. As per the manual:

M.2_1 shares bandwidth with PCIEX16(G5)_2 and PCIEX16(G5)_1. When
M.2_1 is occupied with SSD device, PCIEX16(G5)_2 will be disabled and
PCIEX16(G5)_1 will run x8 only.
